描述:HaiGene的HG miRNA SYBR Green PCR Kit使用Peter改良的miR-Q技术,采用特异性的上下游引物对反转录所得miRNA的cDNA产物进行定量检测【参考文献1】。不同于其它公司的单条特异性引物扩增方法,HaiGene采用双向特异性扩增引物(原理如图1),从而极大提高了miRNA的检测特异性,其可以区分极高相似度的miRNA分子、减少非特异性扩增(图2,3所示)。该试剂盒对可以对任何miRNA分子进行检测。HG miRNA荧光定量PCR试剂盒共有一万余种,试剂盒编号为APXXXXX,每一个miRNA分子对应一个检测试剂盒,可于HG miRNA qPCR kit.xls中查询。如果您研究的mircoRNA分子不在我们的列表中,请来信咨询,我们将及时给您优化设计。该试剂盒中的每一对引物均经过设计优化,确保扩增效率和特异性。使用该方法进行miRNA定量检测已被大量文献所采用【参考文献2~11】。

操作方法

1. 按以下组分配制反转录反应液

microRNA(0.01-0.1 μg)or Total RNA 0.1-2 μg 4×One step miRNA RT Solution 5 μl *10×miRNA RT Primer 2 μl Rnase Free H2O Up to 20 μl

2. 在PCR仪上按以下条件进行反应: 37°C 60min 95°C 5min

3. 获得cDNA产物后使用HG miRNA荧光定量PCR试剂盒进行miRNA定量检测。

根据机型选择步骤A或B,配制反应体系

A: 需要添加ROX染料进行反应孔间信号矫正的Real-Time PCR仪,包括:ABI PRISM7000/7700/7900HT, 7300/7500/7500 Fast Real-Time PCR (Applied

Biosystems);Mx3000P (Stratagene)等。按照如下组分配制20 μl PCR反应体系:

4 进行Real-Time PCR反应, 通常采用两步法,程序如下: Stage 1: 95℃ 15min Stage 2: 95℃ 5s

60℃ 30s 30 ~40cycles Stage 3: Dissociation analysis

5. 扩增结果分析和数据处理

反应结束后确认Real-Time PCR的cDNA样品和NTC阴性对照的扩增曲线和融解曲线。
